Кортиколиберин (corticotropin-releasing factor, CRF) является одним из основных нейрохимических регуляторов ответа организма на стрессирующее воздействие [Dunn, Berridge, 1990; Orth, 1992]. CRF активизирует гипоталамо-гипофизарно-надпочечниковую ось, а также оказывает влияние на негипофизарные структуры. Негормональные эффекты кортиколиберина связаны с модуляцией активности автономной нервной системы и поведенческих реакций. Так, CRF стимулирует работу сердечно-сосудистой системы, влияет на терморегуляцию и метаболизм глюкозы, снижает болевую чувствительность, разнонаправленно изменяет поведение животных в стрессирующих и привычных условиях, а также вызывает активацию биоэлектрической активности мозга вплоть до судорог (в больших дозах) [Brown et al., 1982а; Dunn, Berridge, 1990; Lariviere, Melzack, 2000; Richard et al., 2002].

Кортиколиберин принадлежит к семейству эволюционно древних пептидных гормонов позвоночных, к которому относятся также уротензин I рыб, саувагин амфибий и урокортины I-III млекопитающих [Perrin, Vale, 1999]. Спектры физиологического действия кортиколиберина и родственных ему пептидов в целом сходны и различаются только выраженностью гормональных, вегетативных и поведенческих эффектов [Brown et al., 1982а; Lovejoy, Balment, 1999; Skelton et al., 2000; Richard et al., 2002; Bale, Vale, 2004]. В структуре пептидов сехмейства CRF, состоящих из 3841 аминокислотного остатка, можно выделить гомологичные фрагменты как в С-, так и в N-концевых областях молекул у разных видов и даже классов позвоночных [Lovejoy, Balment, 1999]. Значительное структурное сходство, а также общность физиологического действия пептидов семейства CRF вызывает закономерный интерес и делает актуальным всестороннее изучение их консервативных фрагментов.

Трипептид Pro-Pro-Ile соответствует участку N-концевой области молекул кортиколиберина, уротензина I, саувагина и урокортина I.

В 1989 году Nomizu и Miyamoto, исследуя действие 17 синтетических пептидов, соответствующих различным фрагментам CRF и саувагина, обнаружили, что, подобно полноразмерной молекуле кортиколиберина, CRF4.6 вызывает судороги у мышей при центральном введении. Другие фрагменты CRF были менее активны или подобного действия не оказывали. В отличие от кортиколиберина, CRF4.6 не влиял на секрецию кортикотропина (АКТГ) у крыс [Nomizu, Miyamoto, 1989].

Позже было показано, что трипептид CRF4.6 при интраназальном введении повышает судорожную готовность в модели аудиогенных судорог у крыс [Медведева, 1993]. В меньших дозах CRF4.6 оказывает анксиогенное действие в крестообразном приподнятом лабиринте и потенциирует повышение частоты сердечных сокращений, вызванное сильным звуковым воздействием [Хамидова, 1994; Мартьянов и др., 1994; Martyanov et al., 1994].

Вместе с тем, влияние центрально вводимого трипептида CRF4-6 в диапазоне доз, близких к физиологическим, изучено не было. Несмотря на сходство действия кортиколиберина и его фрагмента CRJF46, нет прямых данных, свидетельствующих о действии этих пептидов на одни и те же рецепторы и анатомические структуры в центральной нервной системе. Проверка этого представляется интересной для понимания структурно-функциональной организации молекулы CRF и родственных пептидов. Кроме того, изучение физиологических эффектов фрагмента CRF46, а также механизмов его действия может быть перспективным для получения новых лекарственных форм коротких пептидов, не вызывающих, в отличие от кортиколиберина, активации эндокринной оси.

Исходя из этого, целью настоящей работы являлось изучение физиологического действия CRF46 при введении в боковые желудочки мозга крыс и участия рецепторов кортиколиберина в реализации эффектов данногоfrтрипептида.

Для достижения указанной цели в работе были поставлены следующие экспериментальные задачи:1. Изучить влияние СИР^б на следующие физиологические параметры:- частоту сердечных сокращений и среднее артериальное давление;- температуру тела;- уровень глюкозы в крови;- болевую чувствительность;- поведение в привычных условиях и стрессирующей обстановке;- электрическую активность головного мозга.

2. Изучить влияние СВД^-б на судорожную активность.

3. Исследовать действие СКР4.6 на артериальное давление, частоту сердечных сокращений, уровень глюкозы в крови, температуру тела и болевую чувствительность после блокады рецепторов кортиколиберина неселективным антагонистом ahCR.F9.41.

4. Оценить влияние полной двусторонней адреналэктомии на глюко- и терморегуляторное действие СЯР4.6.

ВЫВОДЫ

1. CRF4.6 при введении в боковые желудочки головного мозга в диапазоне доз от 1,5 до 150 нмоль/крысу вызывает ряд физиологических изменений:

-повышает артериальное давление и частоту сердечных сокращений у бодрствующих и наркотизированных животных;

- повышает порог болевой чувствительности в тесте «горячая пластина»;

- увеличивает температуру тела у наркотизированных животных;

- повышает уровень глюкозы в крови в тех же условиях;

-активирует поведение крыс животных в привычных условиях (в «домашней клетке»);

- повышает тревожность животных в стрессирующих условиях (в крестообразном приподнятом лабиринте);

-увеличивает электрическую активность головного мозга у бодрствующих животных в условиях свободного поведения.

2. CRF46 при центральном введении в дозах 150-300 нмоль/животное вызывает проявление патологического двигательного возбуждения и индуцирует судорожную активность.

3. Антагонист рецепторов кортиколиберина ahCRF^i блокирует влияние трипептида CRF4.6 на артериальное давление, частоту сердцебиений, температуру тела, уровень глюкозы в крови и болевую чувствительность животных.

4. Полная двусторонняя адреналэктомия не влияет на эффекты трипептида CRF4.6 на температуру тела и уровень глюкозы в крови у наркотизированных крыс.

5. Основываясь на полученных результатах, можно предположить, что трипептидный фрагмент кортиколиберина CRF4.6 является частью одного из активных центров молекулы CRF или его физиологически значимым дериватом, реализация эффектов которого связана с рецепторами кортиколиберина, расположенными вне гипофиза.

ЗАКЛЮЧЕНИЕ

В работе впервые были получены данные о влиянии центрально вводимого трипептидного фрагмента кортиколиберина CRF46 на работу сердечно-сосудистой системы, глюко-, терморегуляцию, болевую чувствительность, поведение животных и активность головного мозга. Было показано, что введение трипептида CRF4.6 в боковые желудочки головного мозга крыс в диапазоне доз, близких к физиологическим (1.5-30 нмоль), увеличивает артериальное давление и частоту сердечных сокращений, повышает уровень глюкозы в крови и температуру тела, снижает болевую чувствительность, стимулирует ориентировочно исследовательское поведение животных в привычной обстановке и угнетает его в новой, стрессирующей ситуации, а также оказывает стимулирующее влияние на биоэлектрическую активность головного мозга крыс. В надфизиологических дозах (150-300 нмоль) CRF4.6 вызывает двигательное возбуждение и индуцирует судорожную активность у крыс.

В целом центральное действие трипептида CRF4.6 соответствует картине , негормональных эффектов родственных кортиколиберину соединений [Dunn, Berridge, 1990; Orth, 1992; Dautzenberg, Hauger, 2002]. Можно предположить, что трипептидный фрагмент является частью одного из активных центров молекулы кортиколиберина или его физиологически значимым эндогенным дериватом.

В работе также было показано, что антагонист рецепторов кортиколиберина ahCRF9.4I блокирует влияние CRF46 на работу сердечнососудистой системы, уровень глюкозы крови, температуру тела, а также болевую чувствительность крыс. Эти данные убедительно доказывают участие рецепторов кортиколиберина или CRF-ергических нейронов, чувствительных к CRF4.6, в реализации физиологического действия трипептида. Кроме того, было продемонстрировано, что двусторонняя адреналэктомия не влияет на развитие гипертермического и гипергликемического эффектов CRF4.6. Можно заключить, что в реализации действия трипептида не участвует гормональная ось, конечным звеном которой является кора надпочечников. Принимая во внимание эти данные, можно предположить, что эффекты трипептида CRF4.fi опосредуются мозговыми рецепторами кортиколиберина, расположенными вне гипофиза или реализуются с участием CRF-epгичeкиx нейронов.
