Актуальность темы

На фоне устойчивого роста интереса к здоровому питанию и экологически ориентированному производству особую актуальность приобретают продукты на растительной основе. Они сочетают длительный срок хранения, минимальную потерю питательных веществ и возможность замены мясных продуктов, что особенно важно для потребителей, снижающих потребление животного белка.

Текстурированные растительные белки представляют собой перспективную основу для таких полуфабрикатов благодаря способности имитировать структуру и вкус мяса. Однако актуальной задачей остаётся расширение их функциональных свойств за счёт включения биологически активных компонентов.

Одним из таких компонентов являются пробиотики — живые микроорганизмы, оказывающие положительное влияние на здоровье человека при достаточном уровне потребления. Наиболее часто используются молочнокислые бактерии и бифидобактерии. Их внедрение в рецептуру требует обеспечения жизнеспособности на всех этапах технологического цикла. Эффективным решением данной задачи является инкапсулирование с использованием защитных матриц на основе альгината, хитозана, желатина и других биополимеров. Применяемые методы — эмульгирование, коацервация, распылительная и сублимационная сушки — подбираются в зависимости от специфики продукта.

Еще одной важной задачей является формирование вкусоароматических характеристик, сопоставимых с мясными изделиями. Одним из решений выступает использование ферментированных биокомпозиций, обеспечивающих летучие соединения, аминокислоты и другие метаболиты, формирующие сложный вкусовой профиль.

Дополнительно, в рамках оценки функциональности компонентов растительного происхождения, был проведён эксперимент по изучению пребиотического потенциала экстракта брокколи как природного стимулятора роста Lactobacillus rhamnosus GG.

Таким образом, при разработке растительных полуфабрикатов, обогащённых пробиотиками, остаются нерешёнными ключевые научно-технологические задачи. К ним относятся обеспечение стабильности и жизнеспособности пробиотических культур в условиях замораживания и хранения, а также создание вкусоароматического профиля, сопоставимого с мясной продукцией. Решение этих проблем требует комплексного подхода к подбору состава, технологии инкапсулирования и интеграции биокомпозиций в структуру продукта.

Цель диссертационной работы - разработка технологии замороженного растительного полуфабриката с функциональной биокомпозицией, полученной ферментированием R. oligosporus и P. shermanii.

На основании поставленной цели были сформулированы следующие задачи:

- Обосновать выбор способа инкапсулирования микроорганизмов для включения в растительные замороженные полуфабрикаты;

- Исследовать выживаемость инкапсулированных пробиотиков после термической обработки и хранении продукта;

- Разработать способ получения биокомпозиции с R. oligosporus и P. shermanii;

- Оценить антимикробную и пребиотическую активность биокомпозиции;

- Разработать технологию производства растительных полуфабрикатов;

- Проанализировать органолептические показатели качества замороженных растительных полуфабрикатов;

- Обосновать сроки годности замороженных и охлажденных растительных полуфабрикатов.

Научная новизна работы

Установлена эффективность способа инкапсулирования пробиотических микроорганизмов методом экструзии в альгинатные микрокапсулы, обеспечивающего сохранение жизнеспособности Lactobacillus rhamnosus GG в замороженных растительных полуфабрикатах после замораживания (до -18 °C) и термической обработки (до 70 °C) на уровне не ниже 106 КОЕ/г.

Обоснована возможность использования инкапсулированного пробиотического штамма Lactobacillus rhamnosus GG сохраняющего жизнеспособность клеток на уровне не менее 76 % после замораживания и термической обработки совместно с функциональной биокомпозицией, полученной последовательной ферментацией R. oligosporus и P. shermanii, формирующей вкусоароматический профиль в технологии замороженных растительных полуфабрикатов.

Установлена антимикробная активность ферментированной R. oligosporus и P. shermanii биокомпозиции по отношению к Bacillus subtilis, а также подтверждена её пребиотическая активность в отношении Lactobacillus rhamnosus GG.

Теоретическая и практическая значимость

Определены параметры ферментации для получения биокомпозиции: ферментация соевой смеси штаммом R. oligosporus при температуре 35 °С в течение 24 ч с последующей ферментацией штаммом P. shermanii при температуре 30 °С в течение 24 ч, обеспечивающие накопление молочной и пропионовой кислот, а также формирование выраженных вкусоароматических свойств готового продукта.

Разработанная рецептура растительных полуфабрикатов обеспечивает содержание пробиотических клеток Lactobacillus rhamnosus GG на уровне не менее 106 КОЕ/г, что соответствует рекомендованному уровню ежедневного потребления пробиотиков.

По результатам исследования микробиологических показателей

безопасности, физико-химических и органолептических показателей качества разработанного продукта установлен его срок годности - 6 мес при температуре -18 °С, в течение которого сохраняется жизнеспособность пробиотиков на уровне не ниже 76 %, стабильность вкусоароматических и сенсорных характеристик на уровне не менее 90 % от исходных значений.

Технология производства ферментированной биокомпозиции на основе изолята соевого белка и микроорганизмов R. oligosporus и P. shermanii прошла апробацию в опытно-промышленных условиях ООО «Нордена».

Основные положения выносимые на защиту

- параметры технологии инкапсулирования клеток пробиотического штамма Lactobacillus rhamnosus GG методом экструзии, обеспечивающие сохранение жизнеспособности микроорганизмов в замороженных растительных полуфабрикатах после термической обработки;

- условия ферментации биокомпозиции с R. oligosporus и P. shermanii;

- технология производства растительных полуфабрикатов с пробиотическими свойствами.

Степень достоверности и апробация результатов работы

Достоверность полученных результатов подтверждается не менее чем трехкратной повторностью получения и обработки данных. Полученные результаты подвергали статистическому анализу в пакете Microsoft Excel (доверительный интервал P = 0,95). В качестве литературных данных использовались статьи, опубликованные в высокорейтинговых журналах, индексируемых ведущими научными базами данных.

ОСНОВНОЕ СОДЕРЖАНИЕ РАБОТЫ

Во введении изложены современное состояние исследуемой проблемы и обоснование её актуальности, сформулированы цель и задачи работы, а также раскрыты теоретическая и практическая значимость, научная новизна исследования и ключевые положения, выносимые на защиту.

В первой главе представлен аналитический обзор отечественных и зарубежных источников, освещающих вопрос применения растительных продуктов в качестве функциональных продуктов питания. Рассмотрены особенности применения текстурированных растительных белков, их роль в формировании структуры и улучшении органолептических свойств растительных продуктов. Проанализированы механизмы действия пробиотиков, их влияние на здоровье человека и эффективность технологических подходов к их введению в состав растительных продуктов. Особое внимание уделено методам инкапсулирования пробиотических микроорганизмов (распылительная сушка, экструзия, эмульгирование, коацервация, сублимационная сушка) для

сохранения жизнеспособности пробиотических микроорганизмов при термической обработке и хранении. Рассмотрены принципы создания биокомпозиции на основе ферментированных растительных субстратов, в том числе влияние ферментации на питательные, антиоксидантные и органолептические свойства продуктов. Отмечена роль микробных сообществ — молочнокислых бактерий, дрожжей и плесневых грибов — в формировании структуры, вкуса и функциональной ценности ферментированных продуктов. Отдельно обсуждена роль комплексного применения технологий текстурирования, ферментации и инкапсулирования для получения растительных функциональных полуфабрикатов с улучшенными потребительскими и биологическими характеристиками.

Объекты настоящего исследования и методы проведения исследования представлены во второй главе. Исследования проводились на базе лабораторий МНЦ «Биотехнологии третьего тысячелетия» факультета биотехнологий Университете ИТМО и «National and Local Joint Engineering Laboratory for Synthesis Transformation and Separation of Extreme Environmental Nutrients» института питания и охраны окружающей среды Харбинского политехнического университета. Объектами исследования являлись: пробиотические культуры микроорганизмов Lacticaseibacillus rhamnosus GG, Rhizopus oligosporus NRLL 2710 и Propionibacterium freudenreichii subsp. shermanii - КМ 186 в виде суспензии и в виде функционального инкапсулированного пищевого ингредиента; растительный полуфабрикат, изготовленный из пшеничного и горохового текстуратов; биокомпозиция, ферментированная с помощью R. oligosporus и P. shermanii;

Представлены результаты комплексных экспериментальных исследований, направленных на разработку технологии растительных полуфабрикатов с функциональной биокомпозицией, содержащих инкапсулированные пробиотические микроорганизмы. Проведено сравнительное изучение методов инкапсулирования, установлен способ,

обеспечивающий высокую степень сохранности клеток LacticaseibacШus rhamnosus GG при термической обработке и низкотемпературном хранении. Определены физико-химические и морфологические характеристики полученных капсул, их термостабильность и выживаемость пробиотических культур при воздействии температуры. Показано, что применение альгинатной матрицы обеспечивает поддержание жизнеспособности микроорганизмов на уровне 59 % после термообработки. Определены параметры ферментации для получения биокомпозиции: ферментация соевой смеси штаммом Я. oligosporus при температуре 35 °С в течение 24 ч с последующей ферментацией штаммом Р. shermanii при температуре 30 °С в течение 24 ч, обеспечивающие накопление молочной и пропионовой кислот, а также формирование выраженных вкусоароматических свойств готового продукта. Дополнительно исследованы пребиотические и антимикробные свойства биокомпозиции. По результатам исследования микробиологических показателей безопасности, физико-химических и органолептических показателей качества разработанного продукта установлен его срок годности - 6 мес при температуре -18 °С.

Разработка биокомпозиции

Для сравнительного анализа эффективности различных подходов к получению функциональных биокомпозиций с заданными свойствами был разработан экспериментальный дизайн, включающий четыре варианта ферментации, различающиеся по типу субстрата, составу микробного консорциума и технологическим параметрам (Таблица А.1).

При получении функциональных биокомпозиций с различными биологически активными свойствами применяли два альтернативных подхода к ферментации растительного сырья: последовательную ферментацию с использованием Я. oligosporus и Р. shermanii на примере с соевой смесью (рисунок 1) и ферментацию с В. subtШs на примере с соевым шротом (рисунок 2).

Для последовательной ферментации подготавливали соевую смесь из изолята соевого белка, 352 г соевого изолята смешивали с 646 мл воды, дополнительно вносили глюкозу (0,5 %), KH2PO4 (0,2 %), MgSO4 (0,05 %), затем смесь стерилизовали в автоклаве при температуре 121 °C в течение 15 мин. Перед ферментацией P. shermanii культивировали в жидкой среде YEL при 30 °C в течение 24 ч в анаэробных условиях, а R. oligosporus культивировали на агаровых пластинах YMP при 35 °C в течение 24 ч; после споруляции споры промывали раствором глицерина (10%, v/v) и Tween80 (0,05%, v/v). После подготовки соевой смеси в конические колбы объёмом 200 мл добавляли 100 мл соевой смеси и 1 мл суспензии спор R. oligosporus. Ферментация проходила в термостате при температуре 35 °C в течение 24 ч, после чего смесь гомогенизировали до 10% сухих веществ и стерилизовали. На втором этапе ферментации в соевую смесь вносили P. shermanii — на 100 мл напитка 1 мл суспензии спор; ферментация проходила в термостате при температуре 30 °C в течение 24 ч, анаэробные условия обеспечивались с помощью эксикатора. На последнем этапе биокомпозицию стерилизовали и хранили при 4 °C. Схема последовательной ферментации биокомпозиции представлена на рисунке 1.

Для ферментации биокомпозиции с В. subtШs использовали 5 г соевого шрота, увлажняли дистиллированной водой в соотношении 1:2 до достижения влажности 67%, затем смесь стерилизовали в автоклаве при температуре 121 °С в течение 15 мин. После охлаждения до комнатной температуры в субстрат добавляли L-лизин (2,5% от массы субстрата) и L-треонин (2,5% от массы субстрата), тщательно перемешивали и выдерживали 30 минут. В. suЫШs N^£7144 культивировали на питательном агаре при 27 °С в течение 24 ч, после чего клетки смывали 5 мл стерильной дистиллированной воды. В конические колбы объёмом 250 мл помещали 5 г подготовленного субстрата и вносили 1 мл бактериальной суспензии В. suЫШs. Ферментация проходила в термостате при

температуре 27 °С в течение 72 ч. После завершения ферментации 20 г ферментированного субстрата помещали в коническую колбу с 250 мл дистиллированной воды, перемешивали на тефлоновой мешалке при 60 °С для дальнейшего детектирования летучих соединений. Схема ферментации биокомпозиции с B. suЫШs представлена на рисунке 2.

Соевая смесь (соевый изолят + вода) выбрана как основной субстрат благодаря высокому содержанию белка (85-90%) и хорошей растворимости. Содержание сухих веществ 35% обеспечивает необходимые реологические свойства для твердофазной ферментации при сохранении достаточной влагоудерживающей способности. Соевый шрот использовался как

альтернативный, экономически более доступный субстрат. Несмотря на более низкое содержание белка (45-48%), шрот содержит значительные количества изофлавонов (даидзеин, генистеин), которые могут быть трансформированы в процессе ферментации в биологически активные агликоны, обладающие выраженными антиоксидантными свойствами [95, 96].

Для первого варианта последовательной ферментации Rhizopus oligosporus NRRL 2710 выбран для первой стадии как продуцент протеолитических ферментов. Данный штамм способен гидролизовать белки сои до свободных аминокислот и коротких пептидов, обеспечивая питательную среду для второй стадии ферментации. Температура культивирования 35°C соответствует оптимуму роста и ферментативной активности R. oligosporus. Propionibacterium freudenreichii subsp. shermanii - КМ 186 используется на второй стадии для синтеза пропионовой кислоты. Выбор данного штамма обусловлен его способностью сбраживать лактат в пропионат [97, 98].

Bacillus subtilis NRCZ144 выбран для третьего и четвертого вариантов как альтернативный продуцент вкусоароматических соединений. Согласно литературным данным [85], штамм способен синтезировать широкий спектр летучих соединений, включая эвкалиптол, пиразины и терпены, формирующие комплексный ароматический профиль. Температура 27°C выбрана как подходящая для проявления ароматобразующей активности штамма при твердофазной ферментации [85].

Глюкоза (0,5%) вносится в первый и второй варианты как легкоусвояемый источник углерода. KH2PO4 (0,2%) выполняет функцию буферной системы, предотвращая резкое закисление среды на начальных этапах ферментации. MgSO4 (0,05%) необходим как кофактор ферментативных реакций, включая активацию протеаз R. oligosporus и ферментов пропионовокислого брожения P. shermanii [97].

L-лизин участвует в образовании пиразинов через реакции Майяра и L-треонин (по 2,5%) служит предшественником альдегидов и эфиров, формирующих комплексный аромат ферментированного продукта.

Представленные варианты ферментации позволяют провести комплексную оценку влияния типа субстрата (соевая смесь и соевый шрот), микробиологического профиля (одностадийная и двухстадийная ферментация) и целевой направленности процесса (синтез органических кислот и формирование ароматических соединений) на функциональные свойства получаемых биокомпозиций. Такой экспериментальный дизайн обеспечивает возможность выбора технологического решения в зависимости от требований к конечному продукту.

Выбор режимов ферментации

Проведена сравнительная характеристика разработанных биокомпозиций с оценкой их технологических и функциональных свойств.

При сравнении вариантов ферментации .№1 и .№2 (Таблица А.1), ключевым отличием является характер субстрата и его подготовка. В варианте №1 использован соевый изолят с содержанием сухих веществ 35%, что обеспечило однородную, легко фильтруемую среду с достаточной буферной ёмкостью и равномерным распределением питательных веществ. Первая фаза ферментации с R. oligosporus проходила в условиях оптимальной консистенции субстрата, что способствовало быстрому росту микромицета. После стерилизации (121 °С, 15 мин) и разведения до 10% сухих веществ среда оставалась стабильной и однородной, подходящей для второй фазы с P. shermanii. Вариант ферментации №2 с соевым шротом обладал схожим составом питательных добавок, но из-за наличия крупнодисперсных нерастворимых волокон и антипитательных факторов наблюдалась неравномерность суспензии, осаждение частиц и локальные дефициты питательных компонентов. Ингибирующее действие антинутриентов приводило к снижению активности ключевых ферментов, а неоднородная среда затрудняла контакт микроорганизмов с субстратом, что

существенно уменьшало выход биомассы и снижало функциональную ценность конечного продукта.

Сопоставление вариантов ферментации №3 и №4 показывает значимость состава и предварительной подготовки субстрата даже при одинаковом виде микроорганизма (В. subШis). Хотя температура ферментации (27 °С, 72 ч) и последующее перемешивание при 60 °С обеспечивали размножение бактерий, ограниченная буферная ёмкость приводила к резким колебаниям рН, замедляя метаболизм и увеличивая лаг-фазу. К тому же этап перемешивания при 60 °С усложнял технологическую схему и повышал энергозатраты. Вариант 4, где в качестве субстрата использован соевый шрот с аналогичным содержанием сухих веществ и теми же аминокислотами, не только сохранял ключевые источники углеводов и белка, но и, благодаря волокнистой структуре шрота, способствовал равномерному удержанию влаги и умеренному высвобождению питательных веществ. Это обеспечивало стабильный рН, равномерное распределение бактерий и устойчивый рост В. suЫШs без излишних технологических операций.

В связи с изложенными ограничениями дальнейшие исследования были сосредоточены на вариантах ферментации № 1 и № 4.

Сравнительный анализ метаболитных профилей биокомпозиций

Для выбора биокомпозиции был проведен сравнительный анализ метаболитных профилей двух вариантов ферментации: биокомпозиции на основе В. subtШs и биокомпозиции последовательной ферментации Я. oligosporus и Р. shermanii. Основными критериями сравнения являлись содержание органических кислот, определяющих вкусовой профиль и консервирующие свойства, а также характер летучих соединений, формирующих аромат продукта.

Биокомпозиция на основе В. subШis

В результате анализа методом ГХ/МС в биокомпозиции фементируемой В. subtШs N^£7144 были идентифицированы следующие основные метаболиты,

представленные в таблице 1.

Алифатическая группа соединений составила 68,63% от общего количества летучих веществ, при этом эвкалиптол являлся доминирующим компонентом, представляя 31,25% всего экстракта и формируя характерный камфорный аромат. Вторым по значимости компонентом стал 3-этил-2,5-диметилпиразин, составивший 26,14%. Бутилгидрокситолуол, относящийся к фенольной группе, составил 15% экстракта и является третьим по содержанию компонентом, обладающим выраженной антиоксидантной активностью. Карриофиллен составил 12,02% от алифатической группы, что соответствует примерно 8,25% от общего содержания, и вносит пряные ароматические ноты. Группа эфиров была представлена альфа-терпинилацетатом (11,3%). Терпинеол из группы спиртов составил 2,55%. Группа кислот была представлена фенилпропандиовой кислотой (0,64%), а соединения кислородсодержащей группы (тауролин и таурултам) составили в сумме 1,85%. Эвкалиптол, как основной компонент, синтезируется при участии борнилдифосфатсинтазы, продуцируемой штаммом B. suЫШs в процессе ферментации соевого жмыха.

Биокомпозиция последовательной ферментации Я. о1що$рогт и Р. ь/ктшапИ

Проведена количественная оценка содержания молочной и пропионовой кислот как основных метаболитов используемых микроорганизмов для сравнительного анализа трёх вариантов ферментации (монокультура Р. яИегтапИ, монокультура Я. о1що$рогт, последовательная ферментация Я. о1що$рогт и Р. зИегтапи) и выбора оптимального биотехнологического процесса. Полученные результаты позволили установить биохимические основы формирования качественных показателей биокомпозиций (рисунок 3).

Молочная кислота, являясь основным продуктом метаболизма бактерий и мицелевых грибов, оказывает влияние на органолептические характеристики и физико-химические свойства ферментированных продуктов. Снижение рН молочной кислотой влияет на текстуру продукта, повышая его густоту и вязкость. В исследуемых образцах содержание молочной кислоты варьировалось от 0,1 г/л до 0,3 г/л (рисунок 3). При избыточном накоплении молочной кислоты

(выше 0,3 г/л) развивается чрезмерная кислотность, что приводит к снижению вкусовых качеств и приемлемости продукта [83].

Пропионовая кислота, продуцируемая P. shermanii, обеспечивает характерный вкус и аромат ферментированных продуктов, а также обладает антимикробным эффектом. Содержание пропионовой кислоты в образцах варьировалось от 0,07 г/л до 0,9 г/л (рисунок 3). Наибольшее содержание (0,9 г/л) было зафиксировано в образце с последовательной ферментацией, что свидетельствует о выраженной активности P. shermanii в данном варианте. Пропионовая кислота в высоких концентрациях снижает активность микрофлоры порчи, повышая срок хранения продукта [83].

Сравнение трех вариантов ферментированных соевых смесей показало, что последовательная ферментация обеспечивала уровни молочной и пропионовой кислот, позволяющие добиться высоких текстурных и вкусовых характеристик. Этот вариант демонстрировал сбалансированное содержание молочной (0,2 г/л) и пропионовой кислот (0,9 г/л) (рисунок 3). В этом варианте наблюдается синергизм между двумя видами микроорганизмов, что способствует не только повышению кислотности и улучшению вкуса, но и продлению срока хранения продукта за счет комбинированного действия молочной и пропионовой кислот. На основании полученных результатов для дальнейших исследований и сравнений была выбрана биокомпозиция последовательной ферментации R. oligosporus и P. shermanii.

Производственных испытаний применения технологии производства ферментированной биокомпозиции на основе изолята соевого белка и микроорганизмов R. oligosporus и Р. shermanii.

Акт составлен главным технологом, к.т.н. ООО «Нордена» Овсюк ЕА. и аспирантом Университета ИТМО Жерняковой A.B. о результатах испытания технологии производства ферментированной биокомпозиции на основе изолята соевого белка и микроорганизмов R. oligosporus и Р. shermanii.

В испытаниях использовались следующие компоненты:

• Изолят соевого белка (3,52 кг)

• Глюкоза (0,5%) . КН2Р04 (0,2%) . MgS04 (0,05%)

• Вода питьевая (СанПиН 2.1.3684-21)

• Микроорганизмы: Rhizopus oligosporus NRLL 2710 и Propionibacterium freudenreichii subsp. shermanii (KM 186)

Все сырье, направляемое для производства, сопровождалось сопроводительной документацией, подтверждающей его качество и безопасность.

Процесс производства биокомпозиции включал следующие этапы:

1. Подготовка соевой смеси:

Для приготовления соевой смеси изолят соевого белка (3,52 кг) растворяли в 6,46 л воды с добавлением глюкозы (0,5%), КН2Р04 (0,2%), и MgS04 (0,05%). Смесь стерилизовали в автоклаве при температуре 121 °С в течение 15 минут. Процесс

стерилизации проводился в промышленной установке с автоматическим контролем температуры и времени.

2. Подготовка культуры микроорганизмов:

о Р. shermanii культивировали в промышленной биореакторной системе в жидкой среде YEL при температуре 30 °С в течение 24 часов в анаэробных условиях с использованием автоматизированной системы контроля газа, о R. oligosporus культивировали на специальных агаровых пластинах YMP в условиях термостатированного инкубатора при температуре 35 °С в течение 24 часов. После этого споры из культуры промывали раствором глицерина (10%, v/v) и Tween 80 (0,05%, v/v) с использованием промышленного оборудования для перераспределения и подготовки спор.

3. Ферментация с участием R. oligosporus:

После подготовки соевой смеси она подавалась на ферментационную установку, где происходила инокуляция R. oligosporus. Для этого на 100 литров соевой смеси добавляли 1 л суспензии спор. Процесс ферментации проводился в промышленных ферментерах при температуре 35 °С в течение 24 часов с контролем микроклимата и показателей pH. После окончания ферментации смесь гомогенизировалась до 10% сухих веществ и стерилизовалась в специализированном стерилизаторе.

4. Ферментация с участием Р. shermanii:

I-Ia втором этапе ферментации в уже подготовленную смесь из соевого белка и R. oligosporus вносили 1 л суспензии спор Р. shermanii на 100 литров. Процесс ферментации проводился в термостатических камерах при температуре 30 °С в течение 24 часов. Анаэробные условия обеспечивались с помощью эксикаторов в условиях закрытого цикла.

5. Завершающая стерилизация и хранение:

На последнем этапе готовая биокомпозиция подвергалась стерилизации в промышленной стерилизационной установке при 121 °С в течение 15 минут. После стерилизации продукт охладился до температуры 4 °С в холодильной системе с автоматизированным контролем температуры, после чего был упакован для хранения и транспортировки.

Производственными испытаниями показано, что использование предложенной технологии позволило:

1. Обеспечить стабильные свойства продукции, что способствует улучшению пищевых характеристик и биологической ценности сосвого белка.

2. Получить органолептические характеристики, соответствующие стандартам качества и безопасности.

Рекомендуется использовать предложенную технологию для производства биокомпозиции полученной ферментированием Я. oligosporus и Р. зИегтапп. Применение биокомпозиции возможно для производства широкого ассортимента пищевой продукции.

ABSTRACT

Improving sasuointestina! flora with probiotics is a promising method of treating and preventing many diseases. However, in food industry1, introduction of probiotics is limited by the shelf life of products, as well as by the reduction of bacterial viability during storage. Thus, introduction of probiotics into foods to be frozen is a promising method to extend the shelf life of probiotic products. However, the storage of living cells in frozen products involves their damage during freezing and subsequent storage at temperatures below 0 C. Therefore, some technological solutions are required to protect probiotics during storage at sub-zero. Today, encapsulation, addition of cryoprotectants and prebiotics. as well as adaptation of microorganisms are widely used techniques of protecting probiotics. The combinauon of various protection methods is a promising way to improve the viability of probiotics during storage. Studies aimed at investigating various combinations of hurdle methods and their effect on bacterial survival are of great interest

This review compiles the most common methods of preserving probiodes in frozen foods, describing their effects, variety of approaches, and effectiveness- The mam mechanisms of bacterial cell damage by freezing are described. Further trends in probiodc protection in frozen foods are also discussed.

1. Introduction

Healthy microflora of gastrointestinal (Gl) tract has a proven positive effect on many systems of human body, fi) It acts as an antagonist for pathogenic microflora, releasing bacteriotoxins; (it) it increases bioavailability of food components, destroying compounds that are not amenable to enzymatic cleavage; (Hi) it synthesizes micronutnenls (amino adds, vitamins); (iv) it improves the absorption of microelements through the intestinal walls; (vj It trains Immune system. Regular consumption of adequate foods including those fortified by proteins, is one of Ihe factors contributing lo healthy microflora.

For a product to be considered probiotic, it must contain an adequate amount of living microorganisms. The number of living bacteria, essential for the product to be considered as a probiotic, varies, but the generally accepted level is 10M08 CFU/g (M„> u i P-.. uv;

of living bacteria can decrease dramatically. Therefore, an Important factor in the production of a probiotic product is the survival of microorganisms during storage (! ripuiJi: ^ Gin -t).

The factors that affect the viability of microorganisms include temperature, humidity, waler activity, osmotic pressure, pH. nutrient composition of the product, and competitive microflora. Low Lernpera-ture storage provides a better viability of microorganisms ( 1 < .101 -i). However, low temperature storage of probiotic products is usually limited by 4-6 weeks (ManoilOvn: el ii.. In tliis regard, the introduction of probiotics into products Lo be frozen and stored at negative temperatures is of great interest Such products Include ice cream, frozen desserts, and frozen semi-finished foods. Frozen products have good prospects In terms of administering functional components (e.g.. probioUcs. vitamins, and mlcronuirients) into the human body. In recent years, microcapsules, nanoemulsions, and oleugels have been increasingly used (o enrich frozen products with functional ingredients. The effects of added components on the consistency, taste, smell, storage capacity of finished products, as well as on their physiological properties arc being actively Studied ( . l\""-). Frozen foods are effective as a reservoir for probioUcs, mainly due lo their storage at negative (below 0 C) temperatures leading to increased viability of microorganisms jilht-r^ di. JlHO). Further research In this direction may be needed to expand the range of frozen probiotic products and lo provide a belter understanding of technological challenges Involved.

On the other hand, production of frozen foods has a number of features to be considered. At room temperature probioUcs In food products are able to proliferate, which helps lo partially compensate for losses of living bacteria during production and storage (Maiv and 1 iir.. .'J'i4)r whereas storage of probiotlcs at sub-zero temperatures leads to the state of suspended animution, which prevents them from replenishing. It makes it necessary to preserve them as much as possible during storage. Frozen storage technology is also hampered by the increased lethality of microorganisms during the freezing process. In addition, once in the human body, probiotic cultures must survive in the hostile environment of the gastrointestinal tract and, having reached the large Intestine, remain active enough to attach to Its epithelium.

In view of the above, it is the vulnerability of probiotic bacteria when introduced into frozen foods that remains a challenge. Thus, the purpose of this review is to summarize and compare the existing techniques for protecting probiotic bacteria introduced into frozen food products and suggest their further development.

2. Methodology

Electronic databases and search engines, including PubMed, Scopus. Science Direct, and Google Scholar up !o April 2022 were searched. Subject Inquiries Included srientiftc articles, reviews, and book chapters on probioUcs, their survival in frozen foods, and techniques to improve their survival. It was only publications in English that were considered. Although most of the 156 publications died are dated later than 2000. there are 21 publications dated before 2000, 4 of which belong to the 1980s, 1 article was published in 1978 and the oldest one dates back 1961, the remaining 15 refer lo the 1990s. The following search terms and their combinations were used: "probiotics'", "frozen food products'', "viability", "encapsulation", 'cryoprotection", 'freezing", "prebiotics', "plant extract".

3. ProbioUcs used today

>'17; TantHKk l HJ7). Bacillus genus (i r<: el jOiD), some yeasts (l.'iiA ti al., JUH>), and other microorganisms ( < m _l*)0) arc also used as probiotics 0 1 !). Lactobacilli are a genus of Gram-posf Uve, non-spore-forming microaerophilic bacteria ('».,.< i > !). Lac-

tobacilli process hexosc with the release of lactic acid. They can convert glucose acung as a homofermentative with the prevailing production of lactic acid, or a heicrofcrmentativc, producing lacUc acid as well as COj, ethanol, and aceUc acid. In the presence of oxygen, the balance is shifted towards the increased release of acetate (llamnic- . " . . . • f ).

Lactobacilli are widely present as a part of dairy products, fermented foods, plants, intestines, and mucous membranes of animals and humans. These bacteria secrete organic adds (]'<,. - v. et -d. __) as wdl as fatty adds, hydrogen peroxide, diacetyl, reuterln, and reuter-icyctin and other products (VanrltnlHr^h, ,; !:-i)), which control the growth of pathogenic microflora.

Lactic acid bacteria arc present in many foods, are part of the normal microflora of the gastrointestinal tract and oral cavity, and have also been used since ancient times for the fermentaUon of foods (Aol-uu \ Marcau, ¿v>5 ).

Bifidobacteria are gram-positive, non-sporc-forming, non-motile anaerobes. They process sugar with the formaUon of lactic and acetic adds (M -i.-jiI -r 31. N95). Bifidobacteria produce several vitamins (Bl, B6, B9. B12, and PP), amino adds (alanine, valine, aspartic add, and threonine), L + lactic add, and also may suppress lactose intolerance (Bh3 ■ ■ -_ui -i '). Despite the GRAS (generally regarded as safe) status of the majority of probiotics, they should be used with caution and GRAS status should be obtained for each specific use (i < >.v, h r-,).

4. Effect of the freezing process on the survival of probiotics

Cells can suffer greater damage during the freezing process than during the storage period (R«inrjdli>v:r:j j : i"! :). This is mainly associated with active ice crystal growth during freezing, which causes medianical damage of the cell membrane. In the freezing process, the probiotlcs cell Is exposed Lo several destructive factors (I iy, ), Lhe most significant of which are the following (DpoJ a h nv H".

1. Thermal stress. 11 leads to denaturation of cell proteins and disturbances in the structure of the cell membrane.

2. Formation of ice crystals. Ice crystals are capable of causing mechanical damage to the cell membrane.

including probiotics. as well as on lipid organization, and membrane fluidity (Balasubruraariuin et al„ 2009).

In addition to the influence of external factors, the type of probiotk culture must also be considered, as different types and even strains react differently to freezing conditions, -'aga: os >;t al. showed a

greater survival rate of Lactobacillus acidophilus La-5 bacteria compared to Bifidobacterium animalis subsp. lactis Bb-12 bacteria, 'lite authors note that the decrease in the number of cultures was observed during freezing and was insignificant during the storage. Lactobacilli, cm the other hand, intensively lost viability in the first 9 weeks, after which they remained relatively stable. Drying techniques are commonly used to extend the shelf life of probiotics ('■ I di. ' ). In particular, freeze drying (lyophili-

sation) of bacteria is widely applied. 'Ihc method consists of freezing the bacterial cells followed by lowering the medium pressure, resulting in sublimation of solvents, bypassing the liquid phase (Hcl1»1\ !'.'• l).The method is Commonly used for the preservation of probiotics in pharmaceuticals. However, reactivation of dried probiotics may require special techniques (Iri Mitu Gin. - Jl4). Moreover, the incorporation of dried probiotics directly into a food product may be associated with the alteration of the organoleptic profile due to cell proliferation and the consequeni accumulation of their metabolites (more relevant for unfrozen products). In addition, the data on the effectiveness of dried probiotics is rather scarce. Therefore, all these aspects need further research.

In addition to crystal growth during freezing, such factors as oxygen exposure and product composition (e.g., acidity (ShuL S if ula. 2(>o0)) may also have a significant effect on the celL Mosl probiotic cultures arc sensitive lo oxygen, which affects the cell both directly and indirectly through free radicals that are released during the oxidation of food components (e.g., fats) (Tnpoih. v _»UM). Selection of appropriate packaging provides effective protection of probiotics from oxidative effects of atmospheric air. Modified packages that include oxygen-scavenging agents are able to protect probiotic cultures (i at: u -•! tiL. 2lw ). Such packages can be used for both refrigerated and frozen products.

Today (here is a variety of hurdle methods to protect bacteria, however, for probiotics m frozen products (he must significant are encapsulation, addition of prebiotics and cryoprotectants, adaptation, and High Pressure Low Temperature freezing method. Each of these methods will be discussed in more detail below.

5.1. Encapsulation

In order to protect cells and prolong their shelf life, encapsulation tcchniquc can be used 0 1 ilfvu ^ i ah, This is a promising

method, which can be used both at room and negative temperatures. Encapsulation is a physico-chemical or mechanical process that involves the incorporation of a target active ingredient inlo a coating that protects it from environmental influences (' lien X 1 > i'} ). Encapsulation prevents die pathogenic effects of competitive microflora, osmotic pressure, oxidative and acidic stress (Qvu & Una. JO') '; Mao .1 ul 1S8S). The capsules can be reservoir-like (form a continuous shell around the encapsulate) or matrix-like (the encapsulate is distributed in matrix volume, including the surface) (Zaidoin X Sbtmonl 2010). Today, there are many different encapsulation techniques that allows one to obtain the desired capsule size (Table ')•

The following materials are used as the capsule shell: (I) proteins (gelatin (amvor •.! al,, 2008), milk proteins (H ..J.-'-di d.. 200'.'), wheat protein (>mi, J al 2010)), (ii) lipids (milk fat 0 J .• J'in ilvj •'! al. 2J22))t (Hi) polysaccharides (alginate (Unwlev -it id., i V"), xanLhan and gellan gum (,'.uo & tirlfl'itiu. 2:100), chitosan (Calbiuiu " al., 2( i! »0, k-carrageenan (Date et ai, 2017), starch and Its derivatives (Ylng ,.•«. aj., 2iML')), and (iv) polymers (polyvinyl alcohol (y«u, K. & Amelnw A.. L 202 i)). 1he mosl common are polysaccharide and protein materials, In particular sodium alginate and carrageenan-based capsules (a£*jo: -a 2019; Chen el ill. 2017).

The combination of different agents and biopolymers can improve cryoprotection properties of the capsules. For example, the comblnaUon of hydrocollolds, such as sodium alginate and methoxyl pectin in certain proportions provides a higher degree of cryoprotection for the probiotic than the usage of these biopolymers alone (Sandoval-Co»Ulltj r\ al 20'i ). Another criterion that affects the survival rate of the encapsulated bacteria is the size of the capsules. For example, Lactoabacillus survives better in frozen storage in alginate capsules larger than 30 pm in size than in capsules with an average diameter of 15 pm (Slui- i Marshall 1 ). When selecting the material for encapsulation, additional conditions should be taken into consideration. For example, if a product containing bacteria in alginate capsules is subjected lo heat treatment, there is a high probability of bacterial death, associated with high porosity of alginate capsules. Possible solutions lo that could be changing the concentration of sodium alginate (4% provides belter stability (Manila! et al . ; IX»)), adding additional biopolymers lo the capsules (Prokul-Ja i' al '¿1121), coating the capsules with an additional layer (l >.-i- • el .J., aoaij MuLarrawi -i u'. 2lXjy), and changing the encapsulation type to reservoir.

The addition of preblotics (fructooligosaccharides (Chen Ch-t. l- a! starch (Sultan • j oL. 204)0), Inulin (Akin aL. 2007;

However, the encapsulation of probiotics has an Impact on the cost of the producL In addition, it is also important to consider possible changes in the organoleptic characteristics of the product ((jur. et id.. 2000).

5.2. Prebiotics

Prebiotics are defined as non-digestible food components that can stimulate the growth and activity of normal intestinal microflora, thus having a positive effect oo host health (01b'jt<n Huberipjid, t'i =;). The addition of prebiotics is a promising technology in probiotic protection. Since the function of prebiotics is often achieved mainly through the addition of probiotic bacterial populations lo the GI tract, the protective effects of prebiotics may be due to synergies in the Interaction between prebiotics and probiotics (synblolics) (In rt aL 2U0'j). Sorting into protective prebiotics is necessary because prebiotics may differ in their ability lo enrich certain groups of probiotic bacleria in a particular area of the stomach (Chen aL, 2017). Akaltn Hid t'rly.r I200M noted that the addition of oligofructose to Lhe ice cream formulation significantly Increased the thermal stability of L. acidophilus La-5 and B. ummulis Bb-12 during freezing. The authors also nole thai oligofructose shows a belter cryoprotective effect compared lo inulin, which is attributed to the positive effect of the latter on the ice cream overrun (increased oxvgenaOon, which reduces the survival of aerophobic bacteria). In addition, oligofructose, having a shorter chain, is easier lo digest for bacleria, thus having a better prebiotic effect. Inulin did not prove lo be a suitable cryoprolcctant when applied at 6.5% and 8.0% (hd i oL 2011).

However, Van a J. 12' 1 reported a positive effect on the survival of L acidophilus and B. lactis when 2% inulin was added lo ice cream, in

addition, the authors analyzed the effect of 15%, 18% and 21% sucrosc concentration in the product on bacterial survival. The best results were obtained with 18% of sucrose and 2% of Inulin.

The introduction of apple, orange, bamboo and oat fiber Into the product slightly reduced the survival of probiotic cultures (Bifidobacterium lactis Bbl2 and Lactobacillus acidophilus) (Akatui et al., 2018). 5.3. Cryoprotcctants

It Is possible to add cryoprotcctants and lyoprotectants to probiotic cultures before freezing. Cryoprotcctants can increase the viscosity of the solution, preventing icc crystal growth by creating a viscous, glassy mass (btmlthdiarya. 2018; SuntlvtiiungUnu ei al., 2008) as well as stabilizing osmotic pressure (Kcts c\ al.. l J'">). In turn, lyoprotectants stabilize the lipid layer of the cell membrane (1 enjiei al. 2i . 9).

Cryoprotcctants are classified into two groups based on their molecular weight. Low molecular weight cryoprotectants are mainly sugars (glucoee (Сшсю et al.. 2015), sucrose (Chnvam et al., 1088; К mm an i •:t о!.. 2011; biatcrbs л al.. 2<KN), trehalose (Nanrr.ani ■-; di.. 2011;

5.4. Adaptation

Adaptation of probiotic cells to storage at reduced temperatures has been reported (Baati -i 2/100). This phenomenon is attributed to a change in the cell membrane composition during cell growth in a modified environment (i.e., cell adaptability) and is traceable to strains of L acidophilus and Pediococcus pentosaceus Cltr. x Dunn, 1997). 5.5. Cryoprotective properties of food ingredients

As stated earlier, the composition of the product has a major Impact on the preservation of probiotics during storage. On tlie other hand, the Ingredients in a food product can also have a protective effect Thus, some food additives used to Impart certain organoleptic or structural properties to the product can protect probiotic bacteria from the effects of negative temperatures and stimulate their growth. For example, some sweeteners such as sugar (. W. -i al. .'t l'CK :i 1 *n & Chen, 2MH.), galactose (Chen Lin. A: Chen. 2fl0<j), trehalose (De oiullo el i! 2005; Ll I and turanose I SI) show

good cryoprotective properties.

Many food stabilizers can have a positive effect on the survival of probiotics during freezing. For example, we have already mentioned the cryoprotective properties of well-known and widely used stabilizers such as xanthan gum. gelatin and alginate. In addition, some stabilizers (pectin, agar and gelatin) have prebiotic properties, providing probiotic microorganisms a substrate for growth.

Plant extracts added to products for various technological purposes can also affect the survival of probiotic bacteria. /Vluuail -i. oi 120211'' showed that 5% apple peel polyphenol extract added In yogurt ice cream resulted in the highest viabdily uf Lactobacillus acidophilus and Bifidobacterium lactis compared to control without extract and lower concentration samples. Although there Is currently little Information on the effect of plant extracts on the viabdity of probiotics in frozen foods, there arc numerous studies of their effect in refrigerated dairy products such as yogurt. These studies demonstrate not only the absence of a negative effect of spices, herbal extracts and essential oils on probiotic cultures, but also an increase in their survivabdity by reducing probiotic oxidative stress (AkU el al. 20! 7; (Jraruilu ul 2.018; llulkun el oL 2022; lllu papalayam m si . 2014; MUuilnovk n j! , 202J; Shchata et al. 2019).

5.6. Higfi Pressure Low Temperature

Pressure shift freezing is also of interest in the preparation of functional products, in particular probiotic enriched products (S .ukuults

6. Discussion

Each of the presented hurdle methods has proven to be an effective means to improve the survival of probiotic bacteria In frozen products. Ihe use of encapsulation requires a careful selection of both the encapsulation method and the appropriate material. As shown In i i :■ .. emulsification and extrusion are the most common encapsulation techniques. The combination of encapsulating materials helps lo increase the percentage of microbial survival during freezing up lo 55% (Zeashun el <d., 2020). The Incorporation of cryoprotectants into the product formulation has also been shown lo be an effective means of protecting probiotics, and In some cases, it has proven to be even better than encapsulation (Di* C.iuil.j <-■! a!. 2005). Since some cryoprotectants are used as direct ingredients in food products, it Is possible lo Introduce diem Into them without any significant Implications for the product flavor. Plant extracts would potentially contribute to the survival of probiutics In frozen products, but this topic has not been developed enough yet and requires further research. As for prebiotics, dieir impact appears to be ambiguous. While symbiotic products have a higher potential than using probiotics alone, prebiotics as a single component lo protect probiotic bacteria during freezing are less effective (up lo 5.8%) than the use of barrier faclors such as cryoprotection and encapsulation. Pre-adaptation of microorganisms lo cold stress may also improve the effectiveness of these barrier methods.

7. Conclusion

Introduction of probiotics in frozen foods is a current technological challenge. Frozen probiotic foods can significantly expand the range of functional food products and make them available and attracuvc for new groups of consumers. However, there are several issues that complicate manufacturing of such products due to the increased lethality of probiotic bacteria during both freezing and frozen storage. They include oxidation of anaerobic bacteria, mechanical damage due to ice crystal growth, osmotic pressure and protein denaturation induced by cold stress. Adjustment of freezing conditions, application of barrier factors, selection of the most psychrophilic microbial species, and their adaptation are used nowadays to meet those challenges. A combination of the existing probiotic protection techniques is a promising approach. For example, adaptation of probiotic cultures could be followed by their incorporation in microcapsules along with cryoprotectant agents. However, this area still needs further research and optimization, lhe use of Ingredients with antioxidant properties including plant extracts to increase the viability of probiotics when frozen docs not seem lo be fairly understood at present. In addition, in vivo studies are required to confirm the positive effect of hurdle techniques for probiotic protection against low temperature stress on the viabUity of microorganisms in Intestinal conditions. Research in these directions is needed in order lo expand the variety of frozen probiotic products and to a better understanding of technological challenges.
