ВВЕДЕНИЕ

Актуальность проблемы. Целлюлоза, гемицеллюлозы, пектиновые полисахариды являются главными компонентами растительной клеточной стенки. Пектиновые полисахариды совместно с гемицеллюлозами образуют матрицу, в которую встроены микрофибриллы целлюлозы. Взаимодействие между полисахаридами обеспечивает не только устойчивые, но и динамичные, гибкие свойства клеточной стенки [1,2].

Среди полисахаридов клеточных стенок высших растений пектиновые вещества (пектиновые полисахариды, пектины) являются на сегодняшний день наиболее сложными и интересными с точки зрения структурной организации и функциональной активности. Пектиновые полисахариды выполняют разнообразные биологические функции в растениях [1] и обладают разноплановой физиологической активностью [3]. Сложная структура пектиновых полисахаридов и наличие большого числа генов в растениях, регулирующих их синтез, свидетельствуют о способности пектинов выполнять разнообразные функции в различные периоды роста и развития растений [4]. Пектиновые полисахариды участвуют в процессе прорастания семян и росте проростков, поддерживают тургор растений, обеспечивают в них водно-солевой обмен, определяют устойчивость растений к засухе и низким температурам, обусловливают резистентность растительной клетки к действию фитопатогенов [1, 5, 6].

В составе пектинов обнаружены такие структурные элементы, как линейный галактуронан (НО), разветвленные ксилогалактуронан, апиогалактуронан, рамногалактуронан-1 (1Ю-1) и рамногалактуронан-П (ЯО-П) [7, 8]. Несмотря на значительные успехи в области структурных исследований полисахаридов, сложность построения и нерегулярный характер углеводных цепей пектиновых макромолекул не позволяют считать их структуру установленной. На сегодняшний день не выяснено, являются ли галактаны, арабинаны и арабино-галактаны отдельными молекулами, сопутствующими пектиновым веществам, или они ковалентно связаны с

пектинами. Кроме того, представления о блочном характере углеводных цепей пектинов и наличии ковалентной связи между структурными элементами в их макромолекулах основаны лишь на косвенных доказательствах и требуют подтверждения. Поэтому в настоящее время нет общепринятой модели строения пектиновых макромолекул [9]. Более того, есть все основания полагать, что не все структурные элементы пектинов установлены и, несомненно, поиск пектинов со строением углеводных цепей, отличных от уже известных, представляет значительный интерес.

Недостаточно охарактеризовано влияние климатических и экологических условий произрастания растений на биосинтез пектинов в клетке. Выявление факторов, влияющих на биосинтез пектинов и на их модификацию, необходимо для определения роли пектинов в онтогенезе растений и, поскольку пектины являются физиологически активными биополимерами, взаимосвязи их структуры и физиологической активности.

Являясь одним из главных компонентов клеточных стенок растений, пектиновые полисахариды входят в состав пищевых волокон (ПВ), составляющих значительную часть растительного пищевого рациона человека. Пектины поступают в организм человека как в составе овощей и фруктов, так и в виде функциональных пищевых ингредиентов и биологически активных пищевых добавок (Б АД). Суточная норма потребления пектинов составляет 5 грамм [10]. Пектины характеризуются многоплановой физиологической активностью и способствуют выведению из организма животных и человека токсинов, солей тяжелых металлов, радионуклидов, обладают иммуномодулирующим и противовоспалительным действием [3]. В основе биологических функций и физиологической активности пектиновых полисахаридов лежит структурная организация их макромолекул.

На сегодняшний день установлена структура пектинов ряда овощей и фруктов [11-31]. Однако спектр употребляемых в пищу растений гораздо шире, а условия экстракции пектиновых полисахаридов, применяемые для их

выделения, значительно отличаются от условий переваривания в желудочно-кишечном тракте (ЖКТ) человека. Более того, структура пектиновых полисахаридов может модифицироваться под действием ряда веществ, секретируемых в ЖКТ. Существенное влияние на структуру пектинов могут оказывать различные факторы: рН, ионная сила, активность ферментов. Сведения об экстракции пектинов из ПВ в условиях гастральной среды и их модификации в процессе пищеварения крайне важны при выявлении структурных элементов, определяющих физиологическую активность пектиновых полисахаридов.

Таким образом, исследование структуры пектинов и ее модификации становится актуальной междисциплинарной задачей, лежащей на стыке биоорганической химии, биохимии и физиологии. Много вопросов, связанных со структурой пектиновых полисахаридов, определяющей функции этих биополимеров в растениях и их физиологическое действие на организм, остаются нерешенными и обусловливают то повышенное внимание, которое уделяется учеными разных специальностей и направлений всестороннему изучению пектиновых полисахаридов.

Цель исследования - структурно-химическое исследование физиологически активных пектиновых полисахаридов и выявление факторов, влияющих на их структуру.

Задачи исследования:

1. Провести скрининг растений европейского Севера России и Монголии на содержание в них пектиновых полисахаридов с целью выявления новых структурных элементов макромолекул пектинов.

2. Провести структурно-химические исследования пектиновых полисахаридов, отличающихся наличием в углеводных цепях значительных участков 1Ю-1, высоким содержанием остатков нейтральных моносахаридов, высокой вязкостью водных растворов и выраженной физиологической активностью.

3. Сравнить пектиновые полисахариды растений, принадлежащих к одному виду, но произрастающих в различных природно-климатических условиях: засушливых резко континентальных и влажных умеренно континентальных, - с целью определения влияния систематического недостатка влаги на структуру синтезируемых ими пектиновых полисахаридов.

4. Оценить воздействие техногенных загрязнений на структуру пектиновых полисахаридов клеточных стенок ряски.

5. Провести структурно-химические исследования пектиновых полисахаридов, экстрагируемых из овощей и фруктов в гастральной среде.

6. Установить влияние гастральной среды на структуру пектиновых полисахаридов.

Основные положения, выносимые на защиту:

1. Галактуронан комарумана, пектина сабельника болотного Сотагит palustre L., содержит линейные и разветвленные участки с 2,4-, 3,4- и 2,3,4-замещенными остатками галактуроновой кислоты в качестве точек разветвлений.

2. В растениях одного вида, а также в растениях разных видов одного рода, независимо от климатических условий их произрастания, синтезируются близкие по моносахаридному составу полисахариды.

3. Систематический недостаток влаги вызывает снижение биосинтеза пектиновых полисахаридов в клеточных стенках растений, не оказывая влияния на их моносахаридный состав.

4. В клеточных стенках ряски малой Lemna minor L. и ряски горбатой L. gibba L. под влиянием техногенных загрязнений изменяется синтез пектинов, в результате чего снижается содержание апиогалактуронана и увеличивается содержание ксилогалактуронана.

5. В условиях гастральной среды из клеточных стенок растений экстрагируются растворимые пектины и частично пектины протопектинового комплекса. Углеводные цепи пектиновых полисахаридов при этом

подвергаются деструкции, степень которой определяется структурными особенностями их макромолекул и повышается с увеличением длительности воздействия гастральной среды.

6. В условиях гастральной среды из клеточных стенок растений совместно с пектиновыми полисахаридами экстрагируется белок, частично связанный с пектиновыми макромолекулами.

Научная новизна.

В составе углеводных цепей пектиновых макромолекул обнаружены фрагменты разветвленного галактуронана, в котором главная и боковые углеводные цепи имеют аналогичное строение и представлены остатками 1,4-связанной а-О-галактопиранозилуроновой кислоты; боковые цепи присоединены к остаткам галактуроновой кислоты главной углеводной цепи галактуронана в С2- или СЗ-положение, при этом небольшое число боковых цепей присоединяется одновременно в С2- и СЗ-положения.

Впервые охарактеризованы пектиновые полисахариды растений, произрастающих в условиях влажного умеренно континентального климата европейского Севера России и сухого резко континентального климата Монголии. Установлено, что в них синтезируются пектиновые полисахариды, имеющие традиционную для пектинов структуру с преимущественным содержанием 1,4-а-0-галактуронана, часть остатков галактуроновой кислоты в котором метилэтерифицирована, и с разветвленной областью, представленной КХЗ-1 с боковыми цепями, образованными остатками галактозы и арабинозы.

Продемонстрировано сходство моносахаридного состава пектиновых полисахаридов растений, принадлежащих к одному роду и произрастающих в различных природно-климатических условиях, и показано, что систематический недостаток влаги вызывает снижение содержания пектиновых полисахаридов в клеточных стенках растений, не оказывая влияния на их структуру.

В условиях гастральной среды из овощей и фруктов экстрагируются растворимые пектиновые полисахариды и пектины протопектинового комплекса совместно с белком, часть которого связана с пектиновыми макромолекулами.

Выявлено, что в условиях гастральной среды пектиновые полисахариды подвергаются деструкции, степень которой зависит от строения их углеводных цепей и времени обработки. Наименьшей деструкции подвергаются пектины, включающие значительные участки галактуронана.

Теоретическая и практическая значимость.

Результаты исследования дополняют и расширяют знания о структуре и биосинтезе пектиновых полисахаридов. Показано, что углеводная цепь галактуронана может иметь как линейный, так и разветвленный характер.

Способность ряски малой L. minor L. и ряски горбатой L. gibba L. реагировать на техногенные загрязнения окружающей среды изменением моносахаридного состава пектиновых полисахаридов позволяет рекомендовать ряску как индикатор чистоты водоемов и прилежащих к ним территорий.

Расширены представления о модификации структуры пектиновых полисахаридов в процессе пищеварения. Полученные данные, свидетельствующие о деструкции углеводных цепей пектинов в условиях гастральной среды, должны учитываться при выявлении структурных элементов пектиновых макромолекул, определяющих их физиологическую активность.

Проведенный обширный скрининг пектинов в растениях существенно дополняет сведения о растительных ресурсах Республики Коми и Монголии, что повышает потенциал их рационального использования в интересах человека. Разработаны технологии получения из растительного сырья иммуностимулирующих и противовоспалительных полисахаридных препаратов. Полученные данные о пектиновых полисахаридах дают

возможность использования их в пищевой промышленности и для создания на их основе новых лечебных и профилактических препаратов для медицины и ветеринарии.

По теме диссертации опубликовано 65 научных работ, в том числе одна монография, четыре патента Российской Федерации, 22 статьи, в том числе 21 статья в рецензируемых журналах, рекомендованных ВАК РФ, из них 11 в зарубежных и 10 статей в российских журналах.

Личный вклад автора в работы, выполненные в соавторстве и включенные в диссертацию, состоял в разработке подходов к исследованию, постановке и решении основных задач, активном участии на всех этапах теоретических и экспериментальных исследований, интерпретации, анализе, систематизации полученных результатов и в их оформлении для публикации.

Структура работы.

Диссертация состоит из введения, трех глав: обзора литературы, обсуждения результатов, экспериментальной части, а также выводов и списка цитируемой литературы, содержащего 380 источников, в том числе 340 на английском языке. Работа изложена на 243 страницах машинописного текста, содержит 30 рисунков и 33 таблицы.

Работа выполнена в Отделе молекулярной иммунологии и биотехнологии Федерального государственного бюджетного учреждения науки Института физиологии Коми научного центра Уральского отделения Российской академии наук в рамках плановых тем НИР «Физиологическая активность полисахаридов в зависимости от структуры (ГР № 01.200 107401) и «Физиологическая активность пектиновых полисахаридов, модифицированных в условиях искусственной гастроэнтеральной среды» (ГР№ 0120.0 602858).

ВЫВОДЫ

1. Показано, что в состав клеточных стенок большинства растений европейского Севера России и Монголии входят пектиновые полисахариды, характеризующиеся преимущественным содержанием линейного 1,4-а-0-галактуронана с частично метилэтерифицированными остатками галактуроновой кислоты и рамногалактуронана-I с боковыми цепями, образованными остатками арабинозы и галактозы.

2. Выявлено, что углеводная цепь комарумана, пектина сабельника болотного Comarum palustre L., в разветвленной области наряду с рамногалактуронаном-I содержит участки разветвленного галактуронана, в котором главная и боковые углеводные цепи образованы 1,4-связанными остатками a-D-галактопиранозилуроновой кислоты. Боковые цепи в разветвленных участках галактуронана присоединены к остаткам галактуроновой кислоты его главной углеводной цепи в С2- или СЗ-положения и одновременно в С2- и СЗ-положения.

3. Найдено, что в растениях одного вида, а также в растениях разных видов одного рода, независимо от климатических условий их произрастания, синтезируются близкие по моносахаридному составу пектиновые полисахариды.

4. Выявлено, что систематический недостаток влаги в период роста растений вызывает снижение содержания пектиновых полисахаридов в клеточных стенках, не оказывая влияния на их моносахаридный состав.

5. В клеточных стенках ряски при произрастании в зоне техногенных загрязнений снижается содержание лемнана, сопровождающееся увеличением в его углеводных цепях количества остатков ксилозы и снижением содержания остатков апиозы.

6. Показано, что в искусственной гастральной среде из овощей и фруктов экстрагируются главным образом растворимые пектины, не связанные с компонентами клеточной стенки, и частично пектиновые полисахариды протопектинового комплекса.

7. Установлено, что в искусственной гастральной среде углеводные цепи пектиновых полисахаридов подвергаются деструкции, степень которой определяется их строением: меньшему расщеплению подвержены пектины, содержащие значительные участки галактуронана. Деструкция углеводных цепей пектинов повышается с увеличением времени их инкубирования в гастральной среде.

8. Совместно с пектинами из овощей и фруктов экстрагируются кислые белки, часть которых связана с пектиновыми макромолекулами. Остатки аспарагиновой и глютаминовой кислот являются главными компонентами соэкстрагирующегося белка.

9. Предложена концепция формирования разнообразия структур пектиновых макромолекул, согласно которой пектиновые полисахариды участвуют в адаптативной перестройке растительной клетки, повышая ее резистентность к воздействию техногенных факторов окружающей среды.
